Cytotoxicity of HA-TCP Scaffold on Human Umbilical Cord Mesenchymal Stem Cells using MTT Assay

Hydroxyapatite-tricalcium phosphate (HA-TCP) scaffold is a three-dimensional structure used to support bone regeneration. Ideally, the should be biocompatible, biodegradable, and non-toxic. The tissue engineering technique uses combined stem cell repair defect. To prove non-toxic properties of scaffold, cytotoxicity test needed for human umbilical cord mesenchymal (HUCMSCs). In this study, 27 samples were divided into eight groups with variant HA-TCP doses ranging from 5–1000 µg. Each treatment group scaffolds was covered in HUCMSCs. added by methyl-thiazol-tetrazolium (MTT) colorimetric assay, counted using an optical density (OD) formula, observed microplate reader. viability cells inverted TMS microscope 100x magnification. MTT assay’s shows that HUCMSCs shed each dose, did not show any toxic effect. higher OD value, viability. It has been found there no significant difference between variable percentage cells.
